2月12日,国际学术期刊Cell Reports(《细胞通讯》)在线发表了中国科学院上海营养与健康研究所隶属的计算生物学研究所(中国科学院-马普学会计算生物学伙伴研究所)韩敬东研究组的论文“Immune cell types and secreted factors contributing tissue inflammation-to-cancer transition and immune therapy response”。该研究基于多组织的癌症基因组学数据构建计算方法,挖掘多个组织中炎症-癌症转变的共同基因模块和影响该过程的关键免疫细胞及相关分泌因子,为癌症早期检测提供了潜在的生物标记物。

尽管人们已知慢性炎症增加多种癌症的风险,然而炎症如何促进癌症发展仍然没有明确。其中,判断炎症组织何时向癌症转变至关重要。为实现无偏检测免疫细胞及其分泌因子影响炎症-癌症转变的分子机制,该课题研究人员开发了计算方法“SwitchDetector”。分别基于肝癌、癌和结肠癌的转录组数据,识别出血管生成作为多种癌症的炎症-癌症转变的共同关键事件;并利用浸润免疫细胞类型的组成情况,以及其分泌或抑制的胞外因子来预测该关键事件的基因表达。这些识别出的胞外因子可作为潜在的癌症前、癌症早期检测的生物标记物,并且可以与血管生成基因一起实现癌症预后和病人免疫治疗响应的准确预测。SwitchDetector软件以及炎症-癌症(inflammation-to-cancer,I2C)数据库在www.inflammation2cancer.org公开发布,可供下载和查询。
