造血系统和神经系统是通过骨髓(BM)壁龛细胞的神经支配而联系起来的。造血干/祖细胞(HSPCs)表达神经递质受体,如g-氨基丁酸(GABA) B型受体亚基1 (GABBR1),提示HSPCs可以受GABA等神经递质直接与GABBR1结合的调控。

Shao等人通过成像质谱分析(IMS),发现内源性GABA分子在BM壁龛中的骨内膜附近区域富集。为了更好的理解GABBR1调节HSPCs的功能,Shao等人构建了一个组成性的Gabbr1敲除的小鼠模型。
 
通过分析发现,与野生型小鼠相比,Gabbr1敲除小鼠的HSPC数量显著减少。而且,在竞争性移植模型中,Gabbr1-/-造血干细胞(HSCs)重建经放射处理小鼠的能力减弱。

无论是在稳定还是应激状态下,Gabbr1-/-造血干细胞的增殖均减弱。克隆试验显示,几乎所有的Gabbr1-/-HSPC都处在一种缓慢或非循环状态。在体外共培养时,Gabbr1-/-HSPC产生的细胞总量少,而且伴有显著的分化缺陷和B细胞系扩增。

为了明确GABBR1激动剂是否可刺激人脐带血(UCB)HSPC,研究人员在将UCB HSPC移植到免疫缺陷小鼠之前,进行了简短的体外治疗,结果显示:与GABBR1拮抗剂或空白治疗相比,GABBR1激动剂治疗的HSPC的长期移植显著增加。

综上所述,本研究结果表明,GABBR1在HSPC增殖中的直接作用,GABBR1或可成为临床移植中改善HSPC移植预后的潜在靶点。
