胞外基质失调以及炎症细胞浸润是牙髓炎发展的特点。性别决定区Y-9 box(SOX9)是一个非常重要的转录因子,它在炎症性关节疾病中受到肿瘤坏死因子α的显著抑制。这篇研究的目的是为了探讨SOX9在牙髓细胞外基质、细胞因子表达和免疫反应中所扮演的角色。研究通过免疫组化、Western blot和定量聚合酶链式反应(qPCR)检测正常和炎症牙髓/人牙髓细胞(HDPCs)中SOX9的表达。使用siRNA SOX9体外抑制牙髓细胞中SOX9的表达;采用qPCR, Western blot和凝胶/胶原酶谱法检测细胞外基质失调,并通过antibody arrays扫面细胞因子的分泌情况。通过细胞迁移测定、细胞粘附测定、吞噬测定和酶联免疫吸附测定检测THP-1的免疫反应情况。通过染色质免疫沉淀测定(ChIP)检测SOX9和其靶基因的相互作用。结果显示,SOX9强表达于正常的牙髓组织及HDPCs内,而在感染牙髓中表达降低。阻断SOX9会抑制I型胶原酶的产生,刺激MMP2和MMP13的酶活性,并调节HDPCs中白细胞介素(IL) 8的产生。阻断SOX9也会显著抑制THP-1的分化和功能活性。ChIP结果显
