SIRT1是NAD +依赖性脱乙酰酶,其在多种细胞和组织中起作用以减轻与年龄相关的疾病。然而,至今为止,人们尚不清楚SIRT1是否也起到预防老化和肌萎缩侧索硬化(ALS)期间运动神经元中累积的病理变化的作用。
最近,来自哈佛医学院和麻省理工学院的研究人员发现,在正常衰老期间,SIRT1的表达在野生型小鼠的脊髓中减少。使用在运动神经元中过度表达或缺乏SIRT1的小鼠模型,研究人员发现,SIRT1减缓了运动神经元在神经肌肉接头(NMJ)处的突触前部位的年龄相关性变性。
当比较正常衰老期间的改变与ALS期间的变化时,脊髓的转录分析显示大于90%的重叠,揭示免疫和炎症反应基因的显着上调和突触转录物的下调。
此外,在运动神经元中过表达SIRT1可以延迟高拷贝SOD1G93A小鼠疾病进展至终末期的时间。
因此,该研究结果表明,ALS与衰老之间存在相似之处,阻止衰老的干预措施也可能减缓这种破坏性疾病的进展。
