NATURE:激活PDGF途径缓解遗传性心脏病

最近,研究人员使用患者特异性诱导的多能干细胞衍生的心肌细胞(iPSC-CM)在体外模拟LMNA相关的DCM

电生理学研究表明,突变体iPSC-CMs表现出异常的钙稳态,导致单细胞水平的心律失常。

从机制上讲,研究人员表明,与同基因对照iPSC-CM相比,血小板衍生生长因子(PDGF)信号通路在突变iPSC-CM中被激活。相反,PDGF信号传导途径的药理学和分子抑制改善了体外突变体iPSC-CM的心律失常表型。

总之,该研究结果表明,PDGF途径的激活参与了LMNA相关DCM的发病机制,并指向PDGF受体PDGFRB)作为潜在的治疗靶标。
