IL-17,一个由Th17产生的关键细胞因子,被认为在哮喘中诱导过敏原特异性Th2细胞激活,嗜酸性粒细胞和嗜中性粒细胞的积累,以及血清免疫球蛋白E的产生。所有这些特征也许在过敏性鼻炎中也起到重要作用。据我们所知,仅有少量研究对利用IL-17拮抗剂来处理过敏性鼻炎的可信性进行过评估。因此,本研究的主要目标是,第一,在过敏性鼻炎小鼠模型中确定Th17和调节性T细胞在鼻粘膜中的地位;第二,探究IL-17对这些细胞的影响和抗IL-17抗体在过敏性鼻炎中的治疗效果抗IL-17抗体在卵清蛋白诱导的过敏性鼻炎BALB/c小鼠激发试验中通过滴鼻方式给药。研究者记录了鼻子摩擦动作和喷嚏的次数,和鼻粘膜中嗜酸性粒细胞和嗜中性粒细胞的水平以及Th1, Th2, Th17,和调节T细胞参数。抗IL-17抗体显著地减少了鼻子摩擦动作和喷嚏的数量,降低了嗜酸性粒细胞和嗜中性粒细胞浸润,减少了Th2和Th17反应,并增强了调节性T细胞反应。抗IL-17抗体处理能有效防护过敏性鼻炎。这些结果促进了我们对过敏性鼻炎发病机制的理解,并且能引导出全新的治疗方法。
