本研究的目的是描述口腔中性粒细胞在健康中显示的炎症表型的形态,并将其与幼稚血液中性粒细胞的形态和慢性牙周炎中的促炎性口腔中性粒细胞进行比较。该形态学的特征在于颗粒含量,吞噬体含量和细胞质以及核变化的差异。我们还检查了在细菌体外刺激的幼稚中性粒细胞和体内全组织样本的口腔中性粒细胞中诱导的形态学变化。

材料与方法:从患有慢性牙周炎和健康个体的患者的血液和唾液样本中分离嗜中性粒细胞。在TEM下观察细胞并在成像软件中分析粒度,细胞质密度,细胞核中的常染色质量和吞噬体含量。将单独的血液中性粒细胞群与口腔链球菌孵育并以相同方式在TEM下分析。从慢性牙周炎患者获得牙龈组织并在TEM下观察,以相同方式分析嗜中性粒细胞。

结果:促炎细胞显示较少的颗粒化,较轻的细胞质和较高量的核常染色质。与炎症性口腔健康中性粒细胞相比,这些变化在促炎性口腔慢性牙周炎中性粒细胞中更为突出。口腔慢性牙周炎中性粒细胞还含有更多的吞噬体,并且含有更多含有未消化细菌的吞噬体。在将它们暴露于S. oralis后,这些变化在幼稚血细胞中部分复制。当在血管中观察时,牙龈组织中的嗜中性粒细胞显示出幼稚的形态,并且当它们穿过结缔组织进入上皮时逐渐显示出促炎形态学变化。

口腔中性粒细胞显示与部分或完全激活一致的形态变化,对应于它们的副炎症或促炎症状态。通过将它们与共生细菌一起孵育,也可以在幼稚细胞中诱导这些变化。当嗜中性粒细胞穿过牙龈组织时,它们的形态变为活化状态。
