PNAS:癌症和邻近正常组织中具有重复不稳定的蛋白质组学和基因组学特征

最近,研究人员开发了一种严格的基于基序的方法,可以直接从蛋白质组学和基因组原始数据中量化重复内容,超微卫星,蛋白质组和基因组中的变异。该方法适用于广泛的肿瘤和正常组织。

研究人员发现,肿瘤中的重复不稳定模式与其患者匹配的相邻正常组织之间具有高度相似性。尽管如此,蛋白质表达和基因组中的肿瘤特异性特征与癌症进展强烈相关,并且强有力地预测致瘤状态。

在患者中,基因组重复不稳定性特征的层次结构准确地重建肿瘤进化,其中原发性肿瘤从转移瘤区分开来。

此外,研究人员观察到患者体内的重复不稳定性和点突变负荷之间的反比关系,与其他体细胞畸变无关。

因此,重复不稳定性是肿瘤进化中的独特,瞬时和补偿性适应性机制,并且是早期检测的潜在信号。
