2017年4月17日,国际学术权威刊物自然出版集团旗下子刊《Nature Nanotechnology》杂志在线发表了美国西雅图福瑞德·哈金森肿瘤研究中心Matthias T. Stephan研究员的一篇研究论文,研究论文报道了研究人员开发出一种可生物降解的纳米颗粒,能在体内编程免疫细胞,使其可以识别和破坏癌细胞。纳米粒子编程后的免疫T细胞,可以快速清除白血病小鼠体内的癌细胞,缓解小鼠病情。

Matthias T. Stephan博士表示,他们的新技术是目前所知的第一个在体内快速编程T细胞,赋予其识别癌细胞能力的技术。重新编程的T细胞可在24小时至48小时内开始工作,使免疫系统能迅速产生足够强的反应,及早杀死癌细胞。

诸多临床试验表明,细胞免疫疗法很有前途,但要广泛用于临床治疗还面临不少挑战。目前进行细胞免疫疗法通常需要几周时间,因为必须要将T细胞从患者体内取出,在特殊的细胞处理设备中培养,进行基因工程改造后,才可再次注入患者体内。而有了新的纳米颗粒,就可以消除这一耗时费钱的步骤,这无疑会推动细胞免疫疗法的应用。

这一可生物降解的纳米颗粒能携带专门针对癌细胞的嵌合抗原受体(CARs)基因,利用分子标记,纳米颗粒能像毛刺一样黏附到T细胞上,被T细胞吞噬到细胞核内并随后溶解。其所携带的CARs基因会被整合到细胞核内的染色体中,使T细胞可以在一两天内对新基因进行解码并产生CAR。几天之内,一支癌细胞“连环杀手”军队即可建成。

小鼠实验表明,使用这种纳米颗粒编程T细胞进行治疗,可使白血病小鼠的平均存活时间从大约两周(中位生存期)提高到58天。

目前这一T细胞编程方法距临床应用还有距离,但斯蒂芬称,他们正在寻求新的方案,使这一基因递送和表达系统能在对人施用时足够安全。他还希望,将来能利用这一方法“向患者提供他们自己身上没有的受体”,用于治疗肝炎、艾滋病等传染病。
