近日,中国科学院长春光学精密机械与物理研究所孙再成研究小组和中国科学院长春应用化学研究所的谢志刚和景遐斌课题组合作开发了基于荧光碳点的诊断治疗纳米药物,为实现个性化治疗奠定了基础,相关成果发表在《先进材料》上(Min Zheng et al. Adv. Mater. DOI: 10.1002/adma. 201306192)。

诊断治疗药物是能够将治疗和诊断功能集成在一起的新型纳米药物,它将成像制剂和治疗药物有机地结合在一起,一次给药后,可以达到诊断和治疗的双重目的,或者实现对治疗效果的跟踪,因而可以提供重要的病理信息,以及肿瘤和患者之间的个体性差异等重要信息。这些信息将有助于医生对治疗策略的制定,包括对治疗时间、药物的选择和剂量等做出明智的决策。这种“个性化治疗”可以提高疗效,降低目标毒性,全面提高患者的生活质量和治愈效果。

作为一种新型的碳纳米材料,荧光碳点所具有的超小的粒径、优异的发光特性、低毒性、良好的生物相容性和亲水性等特征引起了人们极大的研究兴趣。孙再成和谢志刚的研究团队创新性地用多色发光的碳点作为药物载体,通过碳点和抗肿瘤化疗药物奥沙利铂的共价偶联作用合成出新型的超小尺寸(~3nm)诊断治疗纳米药物,这一纳米药物可以同时将成像物质(碳点)和药物释放到特定的位置和器官,实现检测和治疗疾病的一次完成。这种个性化治疗手段既提高了疗效又降低了药物的副作用,从小鼠的肝癌肿瘤的治疗效果来看,该纳米复合药物具有疗效高、毒副作用小的优点。

除此之外,该研究团队还首次利用荧光内滤效应设计了基于碳点的纳米传感器,达到检测水质中常见的无机污染物六价铬离子的目的,该传感器具有成本低、操作简便和分析迅速等优点,相关工作已经发表于ACS Applied Materials & Interfaces(Min Zheng et al. ACS Applied Materials & Interfaces 2013, 5, 13242-13247)。而他们制备的碳点-TiO2复合材料则实现了可见光催化净化水质的特性(Dan Qu et al. Nanoscale 2013, 5, 12272-12277)。

上述研究得到了中国科学院“百人计划”和国家自然科学基金委的支持。
