AUTOPHAGY:我国研究人员发现肿瘤抑制因子有助维持心脏稳态

最近,来自清华北大联合生命中心的研究人员发现,PTENα,首次发现的PTEN亚型,通过促进PARK2募集到受损线粒体来调节线粒体自噬。

PTEN在肿瘤抑制中起重要作用,并且PTEN家族成员参与各种亚细胞位置的多种生物学过程。
最近,来自清华北大联合生命中心的研究人员发现,PTENα,首次发现的PTEN亚型,通过促进PARK2募集到受损线粒体来调节线粒体自噬。研究人员还发现,PTENα缺陷小鼠表现出心脏线粒体的积累,具有结构和功能的异常,并且PTENα缺陷的小鼠心脏更容易受到异丙肾上腺素和缺血再灌注诱导的损伤。通过线粒体自噬的线粒体清除也在PTENα缺陷型心肌细胞中受损。
此外,研究人员发现PTENα与E3泛素连接酶PRKN具有物理相互作用,PRKN是线粒体自噬的重要介质。PTENα通过其N末端的膜结合螺旋结合PRKN,并通过以磷酸酶非依赖性方式增强PRKN自缔合,来促进PRKN线粒体易位。PTENα的缺失会破坏线粒体去极化后PRKN的线粒体易位和线粒体自噬。
研究人员建议PTENα作为维持线粒体功能和心脏稳态的线粒体质量控制器起作用。
