Oncotarget:外泌体miRNA竟然与乳腺癌复发有关!

外泌体(Exosome)是指经由细胞内的囊泡运输,从多泡体排出到细胞外的直径为30-100nm左右的细胞外囊泡。外泌体内部含有蛋白质、脂质、核酸等,被分泌到细胞外后,经由体液被相邻或远离的组织吸收,发挥着细胞间信息传递载体的作用。外泌体miRNA(Exosomal microRNAs)的分析与细胞、组织特异性以及与疾病间的联系引起了研究人员的关注。

研究人员调查了外泌体miRNA在乳腺癌患者血清样品中的预后作用,并比较了血清和肿瘤组织之间的miRNA表达。研究人员通过miRNA PCR芯片分析乳腺癌复发患者(n = 16)与乳腺癌无复发患者(n = 16)的外泌体miRNA谱,并通过qRT-PCR检查乳腺癌复发患者(n = 35)组织和乳腺癌无复发患者(n = 39)组织的miRNA表达情况。

结果显示,在384个miRNA中,乳腺癌复发患者中有3个miRNA(miR-338-3p,miR-340-5p和miR-124-3p)显著上调,8个miRNA(miR-29b-3p,miR-20b-5p,miR-17-5p,miR-130a-3p,miR-18a-5p,miR-195-5p,miR-486-5p和miR-93-5p )显著下调。分析miRNA在肿瘤组织中的表达显示,乳腺癌复发患者在其原发部位和血清中具有较高水平的miR-340,而其肿瘤组织的miR-195-5p, miR-17-5p, miR-93-5p, and miR-130a-3p在其血清中下调,但是这些miRNA在乳腺癌复发患者肿瘤中表达高于乳腺癌无复发患者。Logistic回归分析显示,miR-340-5p,miR-17-5p,miR-130a-3p和miR-93-5p与乳腺癌患者复发显著相关。

该研究表明,miR-340-5p,miR-17-5p,miR-130a-3p和miR-93-5p这几种外泌体miRNA可能是预测乳腺癌复发的有用生物标志物,而miRNA在肿瘤组织和血清之间能够显示出不同的表达模式。这些发现表明,癌细胞可能通过释放外泌体miRNA的选择性机制来调节其自身的进程,这为对于癌细胞的进一步的研究提供参考。
