研究认为,骨髓间充质干细胞可强化结肠癌的放疗效果

放疗是改善结直肠癌(CRC)患者预后的重要方法之一。CRC放疗不仅有助于杀伤癌细胞,而且可通过增强骨髓间充质干细胞(BM-MSC)肿瘤特异性,重建肿瘤的微环境。然而在放疗条件下,原位间充质干细胞以及招募的干细胞发挥的作用尚未被完全阐明。非放疗条件下BM-MSC细胞对肿瘤进展的影响仍存在较大争议:一方面,BM-MSC可调节免疫细胞功能,导致免疫敏感性降低,导致肿瘤复发风险增加;另一方面,它还能分泌多种细胞因子,具有抗癌作用。

研究人员诱导hBM-MSC细胞在放疗后分泌细胞因子,进行ELISA和流式细胞检测,采用AutoMac分离技术分离hBM-MSC与CRC细胞,共培养组和对照组细胞分别用紫外线灯和x射线照射后进行增殖试验和存活率测定。

研究发现体外BM-MSC细胞可诱导CRC细胞的上皮细胞-间充质转化。低剂量紫外线灯和x射线辐射条件下,BM-MSC通过分泌细胞因子(TNF-α和IFN-γ)表现出抗肿瘤效果,抑制CRC细胞增殖及诱导细胞凋亡。辐射可诱导caspase 3断裂、抑制细胞内磷脂酰肌醇3激酶(PI3K)/AKT和细胞外信号调节激酶的磷酸化,上述信号途径有助于加速癌细胞死亡。

研究认为,骨髓间充质干细胞可强化结肠癌的放疗效果。
