SCI ADV:依诺沙星可以帮助减肥?

肥胖症正影响着全世界6亿多人,并导致2型糖尿病、心血管疾病和癌症的发生几率变高。肥胖是食物摄入和能量消耗之间不平衡的结果,导致多余的能量以脂质的形式在脂肪细胞内积聚。

骨骼肌和脂肪是体内能量消耗的主要组织。为了给氧化代谢提供燃料和产生5′-三磷酸腺苷(ATP),骨骼肌使用大量从饮食中获得的或由白色脂肪组织(WAT)提供的底物,特别是在运动时。热量是这些过程的副产品,也可以通过诱导未耦合的呼吸和无效循环来促进热量产生,这反过来又有助于哺乳动物的体温控制,以及棕色脂肪组织(BAT)诱导的产热。

微RNAs(miRNAs)已被报导参与氧化代谢和棕色脂肪细胞的身份特征。在这里,研究人员测试了小分子依诺沙星治疗导致的miRNA表达的广泛变化是否会诱导脂肪棕色化并防止肥胖。

结果显示,依诺沙星减轻了饮食诱导的小鼠肥胖,这与能量消耗增加有关。一致地,恩诺沙星处理小鼠的皮下白色和棕色脂肪组织以及骨骼肌具有较高的与产热和氧化代谢相关的标志物水平。这些效应是细胞自主的,因为它们在体外的小鼠和人类细胞模型中都能重现。

在脂肪前体细胞中,依诺沙星导致miR-34a-5p表达减少,其靶基因(如Fgfr1、Klb和Sirt1)上调,从而增加FGF21信号传导,促进棕色脂肪生成。

因此,这些数据表明,依诺沙星通过促进产热信号传导,以及诱导脂肪组织和骨骼肌的氧化代谢来对抗肥胖,其机制至少部分涉及miRNA介导的调节。
