抗坏血酸磷酸酯钠(SAP)是一种亲水性稳定的L-抗坏血酸衍生物,细胞磷酸酶将其水解为游离的抗坏血酸(AA),释放在培养基中。AA参与牙周组织的稳定和愈合,具有调节成骨细胞活性作用。

抗坏血酸磷酸酯钠(SAP)是一种亲水性稳定的L-抗坏血酸衍生物,细胞磷酸酶将其水解为游离的抗坏血酸(AA),释放在培养基中。AA参与牙周组织的稳定和愈合,具有调节成骨细胞活性的作用,刺激胶原蛋白细胞外基质的沉积,并诱导成骨细胞表型相关的基因。它还具有消除牙周病中防御细胞大量产生的活性氧的作用。本研究的旨在评估SAP对成骨细胞活性和分化的作用。

本实验采用MC3T3-E1株的小鼠成骨细胞前体细胞。通过台盼蓝染料排除法检测细胞活力,定量PCR评估成骨细胞分化相关基因的表达。采用ELISA法评估胶原蛋白I的分泌情况,并用茜素红S染色法测定矿化基质的形成。

结果表明,在50-500μmol/L的浓度下,SAP不影响成骨前体细胞的活力,但诱导RUNX2,COL1A1和BGLAP2,胶原蛋白I的分泌水平增高,促进细胞分化;与对照组相比,在200或400μmol/L的浓度下,细胞的细胞外基质矿化程度增加。

本实验检测了SAP诱导成骨前体细胞分化的稳定性和能力。将SAP结合到局部释放装置、膜/支架或生物材料中,可能会促进骨组织的形成,从而促进牙周愈合。
