PNAS:高尔基体异常是神经退行性病变关键原因之一

100多年前,意大利细胞学家Camillo Golgi在小脑浦肯野(Purkinje)细胞中发现高尔基体,现已证明高尔基体是细胞分泌途径的核心细胞器。近年来发现高尔基体的结构紊乱广泛存在于神经退行性疾病中。但是,高尔基体结构异常是不是造成神经退行性疾病的原因,一直是悬而未决的科学问题。中国科学院遗传与发育生物学研究所鲍时来研究组为了揭示高尔基体高级结构的生理学功能,构建了全身和多个器官特异性敲除高


100多年前,意大利细胞学家Camillo Golgi在小脑浦肯野(Purkinje)细胞中发现高尔基体,现已证明高尔基体是细胞分泌途径的核心细胞器。近年来发现高尔基体的结构紊乱广泛存在于神经退行性疾病中。但是,高尔基体结构异常是不是造成神经退行性疾病的原因,一直是悬而未决的科学问题。

中国科学院遗传与发育生物学研究所鲍时来研究组为了揭示高尔基体高级结构的生理学功能,构建了全身和多个器官特异性敲除高尔基体基质蛋白GM130基因的小鼠,发现在神经细胞特异性敲除GM130破坏高尔基体高级结构,导致小鼠发育迟缓,并出现退行性的运动障碍;而且发现这种退行性运动障碍是由于浦肯野细胞的进行性死亡,小脑渐渐萎缩等神经系统缺陷导致的;GM130缺失虽然在发育早期不影响高尔基体在浦肯野细胞中的极性形成,但是由于阻碍了高尔基体与中心体的连接,使得发育后期高尔基体极性分布的维持能力丧失,降低了细胞内的物质运输功能,从而导致神经元的退行性病变。

该研究首次证明了高尔基体的结构和功能的紊乱是神经退行性疾病发生的重要起因,并解释了当年高尔基能够在浦肯野细胞中发现高尔基体的原因。


该研究于2016年12月27日在线发表于《美国科学院院刊》(PNAS,DOI:10.1073/pnas.1608576114),并作为封面文章于2017年1月10日正式发表。鲍时来研究组的博士生刘春宜为该文第一作者,助理研究员梅玫和博士李秋伶是该论文的共同第一作者。英国曼彻斯特大学教授Martin Lowe和中科院动物研究所研究员高飞参与了其中的研究工作。该研究工作得到了国家自然科学基金委、中科院和英国生物技术和生物科学研究委员会(BBSRC)的资助。
