根尖牙乳头来源的干细胞(SCAP)可能会受到未发育完成坏死牙齿感染髓腔内因脂多糖(LPS)激活而释放的炎症介质的影响。因此,这篇研究的目的是为了评估肾素-血管紧张素系统(RAS)的存在与否以及血管紧张素(Ang II)对体外培养的SCAP的调控作用。

研究将原代培养的SCAP施以LPS (0.1-10 μg/mL)刺激以检测细胞活性并定量检测趋化因子CCL2的表达。RAS成分分析通过检测血管紧张素原(AGTN), 肽基二肽酶(ACE), 肾素, 血管紧张素受体1 (AT1)和2 (AT2)以及Mas受体的基因表达实现。通过SCAP悬浮物检测Ang II。采用免疫荧光检测AGTN和AT1。随后,细胞施以Ang II刺激,检测活性/增殖情况并定量分析CCL2和IL-6以及矿化情况。所得数据通过analysis of variance以及Tukey post hoc comparisons或 Student t检验进行统计学评估。P values <.05被认为具有显著性。

结果显示,LPS在1和10 μg/mL的浓度时会增加CCL2表达。可检测到AGTN, renin, ACE和AT1的表达,但仅有ACE是由LPS刺激而表达上调。Ang II肽可在SCAP悬浮物中检测到,但并不会因LPS刺激而改变。免疫染色可检测到AGTN和AT1的表达。Ang II能够显著诱导SCAP增殖,增加CCL2表达,下调IL-6的释放并降低SCAP的矿化率。

结论:根尖牙乳头存在局部的RAS,并且Ang II能够体外调节SCAP的功能。
