在胚胎发育中,干细胞特化受到了严格的调控。蛋白复合体PRC2就是一个主要的调控途径,确保细胞在正确的时间启动正确的基因。此前研究显示,PRC2控制基因活性是果蝇和小鼠发育所必需的。但人们还不清楚PRC2在人类发育中扮演着什么样的角色。

研究人员用CRISPR基因编辑技术从人类胚胎干细胞删除PRC2。缺乏PRC2导致细胞启动了许多平时不活跃的基因。有趣的是,这组基因在胚胎发育形成特化细胞的时候有重要作用。由此可见,在人类发育初期使这些特化基因保持关闭是PRC2的一个主要功能。研究显示,当这组基因异常启动的时候,胚胎干细胞的质量和稳定性都会受到影响。这样的改变导致缺乏PRC2的胚胎干细胞无法正确特化为成熟细胞。
