Biomaterials:研究揭示充质干细胞外泌体修复骨软骨缺损的机制

结果显示,外泌体介导的骨软骨缺损修复的特点是增加了细胞的增殖和浸润,增强基质合成和再生的免疫表型。通过软骨细胞培养,瓦米提介导的软骨修复可以促进细胞的快速增殖和浸润,其原因是外泌体介导胞外体CD73介导腺苷激活Akt和ERK信号转导。Akt和ERK磷酸化抑制剂可抑制外泌体介导的细胞的增殖和迁移,但不影响基质的合成。

通过AMPCP衰减Akt和ERK信号通路可证实外泌体CD73的作用。外泌体处理的骨缺损也显示一个再生的免疫表型,其特征是CD163 +再生M2巨噬细胞浸润多于CD86+ M1型巨噬细胞,伴随着促炎症滑膜细胞因子IL-1和TNF-α的相应减少。

综上所述,该研究结果表明,MSC外泌体有效的骨软骨再生作用是通过协调调动多种类型的细胞和多种细胞过程激活而发挥的。
