Nature:清除衰老细胞可以有效延长小鼠寿命

论文通信作者、梅奥诊所生物化学与分子生物学主席Jan van Deursen博士说,“细胞衰老是遭受损伤的细胞用来阻止细胞分裂的起着紧急刹车作用的生物学机制。鉴于阻止这些细胞发生细胞分裂在预防癌症中发挥着重要作用,人们在理论上就认为一旦启动紧急刹车,这些细胞将不再是必需的。”

在这项研究中,梅约诊所研究人员利用一种转基因技术允许药物诱导衰老细胞从正常小鼠体内清除。一旦注射一种被称作AP20187的药物将衰老细胞移除,就会延迟肿瘤形成,并降低几个器官发生年龄相关性退化。接受注射的小鼠平均寿命延长了17%到35%。他们还证实这些小鼠的脂肪组织、肌肉组织和肾脏组织看起来更加健康,而且炎症下降。

免疫系统会定期清除这些衰老细胞,但是随着时间的推移也变得不那么有效。衰老细胞产生损害附近细胞和导致慢性炎症的因子,而慢性炎症与虚弱和年龄相关性疾病密切相关。

van Deursen博士认为,伴随衰老而积累的衰老细胞是非常不好的,会损害人的器官和组织,因而会缩短人的寿命,以及健康寿命(healthspan)。鉴于在小鼠体内能够清除这些细胞同时不会产生副作用,这似乎表明能够清除衰老细胞的药物或其他化合物可能有助于治疗年龄相关性残疾、疾病或症状。

论文第一作者Darren Baker博士也对这项研究对人类的潜在影响感到乐观。

Baker博士说,“清除衰老细胞的优势在于仅需清除60%~70%的衰老细胞就能够产生显著性的治疗效果。如果能够应用于临床,那么鉴于衰老细胞不能快速增值,利用一种药物高效地和快速地清除足够多的衰老细胞将对健康寿命和总寿命产生深刻影响。”
