Eur Heart J:脂肪细胞脂肪酸结合蛋白通过破坏血脑屏障加剧脑缺血

A-FABP是脑缺血损伤的关键介导因素,可促进MMP-9介导的BBB破坏。抑制A-FABP是改善IS预后的潜在策略。

脂肪细胞脂肪酸结合蛋白(A-FABP)是一种参与多种代谢疾病的脂肪因子。缺血性脑卒中(IS)患者循环中A-FABP水平升高与不良预后成正相关。没有研究探究A-FABP在IS发病机理中的作用。

近日,心血管领域权威杂志Eur Heart J上发表了一篇研究文章,研究人员旨在明确A-FABP是否介导血脑屏障(BBB)破坏,如果是,则探讨这种有害作用的分子机制。

大脑中动脉闭塞小鼠循环中的A-FABP水平及其脑中表达增加。A-FABP的基因敲除和药理抑制作用减轻了梗死体积、脑水肿、神经功能缺损和神经元凋亡,减轻了脑缺血损伤。 BBB破坏有所减轻,并伴随着紧密连接蛋白的降解和基质金属蛋白酶9(MMP-9)减少。在急性IS患者中,增加的循环A-FABP水平与MMP-9和脑梗死体积呈正相关。从机制上讲,局部缺血诱导了外周血单核细胞衍生巨噬细胞和脑中小胶质细胞A-FABP增加,其可通过增强JNK / c-Jun信号传导促进了MMP-9反式激活,从而增强了紧密连接蛋白的降解和BBB的泄漏。MMP-9的药理抑制作用可预防A-FABP的有害作用。

由此可见,A-FABP是脑缺血损伤的关键介导因素,可促进MMP-9介导的BBB破坏。抑制A-FABP是改善IS预后的潜在策略。
