AGING CELL:脂肪组织来源的基质细胞有助于衰老过程中的血管重塑

衰老是血管疾病的独立危险因素。血管周围脂肪组织(PVAT)是血管系统的活性成分,在衰老过程中导致血管功能障碍。研究人员认为,鉴定潜在细胞类型及其在衰老过程中的变化可以提供关于衰老相关心血管疾病的临床相关性的有意义的见解。

最近,研究人员利用单细胞RNA序列来表征PVAT(PVASCs)中的驻留基质细胞,并鉴定年轻和老年PVASC之间的不同点。

生物信息学分析显示,在衰老过程中,PVASCs的内皮和棕色脂肪形成分化能力下降,这导致周围血管输送至结扎颈动脉后的新内膜增生。

原理上说,体外和体内研究均表明衰老诱导的过氧化物酶体增殖物激活受体-γ共激活因子-1α(PGC1α)的丧失是衰老PVASC中棕色脂肪形成分化减少的关键调节因子。

研究人员进一步证明了人PVASCs(hPVASCs)的存在,以及PGC1α的过表达改善了hPVASC递送诱导的血管重塑。

该研究结果强调PVASCs的分化能力在老化过程中发生变化,老年PVASCs中PGC1α的缺失通过减少棕色脂肪的形成分化,促进血管重塑。
