Sci Rep:Bretschneider溶液诱导心脏手术患者的尿液代谢组改变

Bretschneider组氨酸-色氨酸-酮戊二酸(HTK)心脏停搏液的出现代表着心脏手术的重大进步,该停搏液具有显著的心肌保护作用。然而,这种添加物在全身引起的代谢变化尚未得到系统研究。这项研究使用基于非靶向质谱法深入探索尿液代谢组,试图整体和系统地描述HTK患者的代谢紊乱。

从100名接受过心脏手术的患者中前瞻性收集尿液样本,并使用高效化学同位素标记液相色谱-质谱法(LC-MS)对代谢组学变化进行分析。使用靶向来自尿液样品胺/酚代谢物的差异13C-/12C-丹酰标记LC-MS,对14,642个峰对或代谢物进行定量。

共确定了223种代谢物有显著的浓度变化(倍数变化>5),并聚集了几种来自这些失调代谢物的潜在代谢途径图。研究数据表明接受HTK的患者中,组氨酸代谢上调,随后伴谷氨酰胺/谷氨酸代谢增加、嘌呤和嘧啶代谢改变、维生素B6代谢增强。

这项研究提供了确凿证据表明,HTK溶液在几种代谢途径引起明显的紊乱,并为进一步研究HTK的器官保护或潜在害处的关键机制奠定了基础。
