研究者Erwin Bottinger博士表示,我们的目标就是阐明内皮细胞的线粒体氧化应激反应调节从血液到尿液的蛋白通过的分子机制,以及肾脏如何过滤那些废弃物的机理;相关成果为研究者开发治疗慢性肾病的新型靶向疗法提供了一定的希望和思路。

文章中,研究者使用小鼠模型,诱导其在肾脏过滤部位结疤,这就会促使蛋白质进入尿液,从而干扰肾脏中废弃物的清除;同时研究者也检测了不同的信号机制以及细胞反应的工作原理。肾小球过滤屏障的关键细胞足细胞可以修饰内皮缩血管肽-1,从而激活内皮素A型受体,这种内皮素A型受体的激活往往会随着内皮细胞线粒体氧化应激反应而出现一定的障碍表现。

研究者Bottinger表示,以线粒体和内皮缩血管肽为靶点的抗氧化剂往往会改变其抑制细胞消耗和肾脏中疤痕形成的模式,目前我们急需要开发出治疗慢性肾病的新型疗法;而肾素-血管紧张素抑制剂和血管紧张素受体抑制剂目前可以广泛有效地用于抑制肾脏衰竭,我们需要更多有效的药物来治疗身患慢性肾病的个体,本文的研究也为开发治疗慢性肾病的靶向疗法提供了一定的帮助。
