J Endod:硅酸钙类封闭剂在2和3维细胞培养模型中产生的细胞因子和毒性研究

这篇研究的目的是为了评估硅酸钙类封闭剂对人牙周膜干细胞(PDLSCs)细胞因子的产生以及活性作用。

这篇研究的目的是为了评估硅酸钙类封闭剂(ie, BioRoot RCS [Septodont, Saint Maur des Fosses, France], ProRoot ES [Dentsply Sirona, York, PA]和MTA Fillapex [Angelus, Londrina, PR, Brazil])对人牙周膜干细胞(PDLSCs)细胞因子的产生以及活性作用。AH Plus (Dentsply DeTrey GmbH, Konstanz, Germany)作为对照材料。

研究将PDLSCs培养于含牙髓封闭剂洗脱物的2维或3维条件(in 0.15%-0.5% PuraMatrix [BD Biosciences, Bedford, MA])下24小时。此外,通过体外牙根模型实验程序评估牙髓封闭剂洗脱物的细胞毒性。通过3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide assay检测PDLSCs活性。通过HCYTMAG-60K-PX41 Milliplex kit (EMD Millipore, Burlington, MA)检测PDLSCs培养基中细胞因子的含量(interleukin [IL]-6, IL-8, growth-regulated oncogene, IL,-4和IL-10)。

结果显示,在2维培养条件下,BioRoot RCS展现出PDLSC良好的生存力。不论稀释度如何,ProRoot ES对PDLSC活力无影响。即使在最低的提取浓度(1:1, 1:2和1:4),MTA Fillapex均表现出较强的细胞毒性。PuraMatrix中PDLSCs的包裹趋于减少封闭剂的毒性作用。在体外3维牙根模型实验过程中,BioRoot RCS, ProRoot ES和MTA Fillapex展现出细胞相容性模式。不同的硅酸钙类封闭剂产出不同量的促炎症因子。BioRoot RCS刺激PDLSCs释放大量的IL-10以及较少程度的IL-4 (P < .05)。

结论:BioRoot RCS和ProRoot ES不会诱导PDLSCs释放促炎症因子并且会促进抗炎症因子的分泌,它可能会减弱最初的炎症反应从而对局部产生积极的影响。
