复制危机是一种与衰老无关的过程,通过消除细胞周期检查点中断的癌前细胞,作为避免致癌转化的最终屏障。它起着有效的肿瘤抑制剂的作用,最终导致广泛的细胞死亡。细胞很少逃避消除并向恶性肿瘤发展,但危机中细胞死亡的机制尚不清楚。

最近,研究人员表明,巨自噬在危机期间成纤维细胞和上皮细胞的死亡中起主导作用。自噬的激活对于细胞死亡是至关重要的,因为其抑制促进了危机的绕过,持续的增殖和基因组不稳定性的积累。

端粒功能障碍特异性地触发自噬,暗示端粒驱动的自噬途径不是由染色体内断裂引起的。端粒DNA损伤产生具有脆弱核包膜的细胞溶质DNA种类,其经历自发破坏。细胞溶质染色质片段激活cGAS-STING(干扰素基因的环状GMP-AMP合酶刺激物)途径并参与自噬机制。

因此,这些数据表明,自噬是肿瘤抑制机制的一个组成部分,并且自噬功能的丧失是癌症发生所必需的。
