基于活病毒采用突变和高通量筛选技术快速开发流感病毒疫苗

基于减活性病毒的疫苗（live attenuated vaccine）能够产生更强的更持久的体液免疫（humoral immunity）和细胞免疫(cellular immunity)反应。但是减活病毒的过程，以及筛选具有保护作用的减活病毒需要一定的时间。减活病毒疫苗的安全性也是一个重要问题。

在新一期的Cell Host & Microbe杂志中，Lulan Wang及其同事基于活流感病毒，运用全基因组诱导突变技术，以及一种高效筛选流程，筛选出一种减活性的流感病毒疫苗W7-791。该种疫苗具有很高的安全性。即使是使用其母病毒的50%致死剂量（LD50）的100倍，这种减活的疫苗在成年或者新生实验鼠中也只产生暂时的轻微感染症状。单次接种此种减活病毒就能在实验鼠上彻底保护H1N1,H3N2以及H5N1流感的感染，并且在实验雪貂上加速病毒的清除。从接种疫苗后的实验鼠身上分离的T细胞，再移植这些T细胞到接受的实验鼠，也被证明这些移植的T细胞有更强的抵抗流感病毒的能力。

此项研究证明了对于活病毒运用突变技术快速建立减活病毒库，然后运用高通量筛选系统发现能够对抗病毒的潜在疫苗，从而开发疫苗的可行性和理论基础。对于突发的流行性病毒感染的应对及疫苗开发，给出了重要的可行方案。
