Diabetologia:一项关于人类胰岛移植和血管重建的纵向成像研究

快速而充分的胰岛重建和胰岛-细胞外基质(ECM)相互作用的恢复是影响1糖尿病患者胰岛存活和移植胰岛功能的重要因素。由于包囊胰岛的ECM在胰岛分离过程中降解,因此了解移植后的血管再生和移植后的定植过程至关重要,需要进一步的研究。

在此,我们采用纵向和高分辨率成像方法来研究移植后11个月胰岛移植的动态过程。将人和小鼠胰岛移植入小鼠眼球前房,使用NOD.ROSA-tomato.Rag2−/− or B6.ROSA-tomato研究宿主细胞与供体细胞的扩增,以及宿主细胞对促进移植物包封和血管化等方面的作用。

宿主来源的成纤维细胞样基质细胞群迅速迁移到被移植的胰岛周围,并帮助形成基底膜样结构。此外,我们发现血管网络虽然由宿主内皮细胞重建,但仍然保留了供体胰岛的整体结构。

研究结果表明,在这种移植情况下,纤维母细胞样基质细胞似乎成为ECM的主要生产者,或作为支架,使其他产生ECM的细胞重建胰岛周围样基底膜。这可能对我们理解长期移植排斥反应和设计干预这一过程的新策略具有意义。
