Lancet:人神经干细胞产品(CTX-DP)或将为卒中患者带来希望

研究人员开展了一项开放性、单一地点、剂量递增的研究。年龄在60岁以上,有稳定的残疾(国立卫生研究院卒中量表[NIHSS]评分≥6和改良Rankin量表评分2-4),且在缺血性卒中后6-60个月的男性通过立体定向注射同侧壳核被植入干细胞,单剂量为200万,500万, 1000万,或2000万个细胞。临床和脑成像数据在研究超过2年时进行收集。主要终点是安全性(不良事件和神经系统的变化)。

在2010年9月和2013年1月期间,13名男性被招募到此研究中,其中11人(平均年龄69岁,范围60-82)接受了植入CTX-DP。植入前平均NIHSS评分为7(IQR 6-8),从卒中发生到研究开始的平均时间为29(SD 14)个月。其中3名男性仅有皮质下梗死,7名男性有右大脑半球梗死。研究过程中没有发生免疫学或细胞相关的不良事件。其他不良事件与该过程或患者的合并症有关。5名患者的液体衰减反转恢复序列T2加权成像发现在注射区域有高强度信号。在2年时,NIHSS评分改善为0-5(平均数为2)个点之间。

单脑内剂量高达20万个细胞的CTX-DP并没有诱导不良事件,并且患者的神经功能也有所改善。该研究支持在卒中患者中进一步调查CTX-DP的效果。
