J Endod:2种生物陶瓷类封闭剂生物相容性和成骨潜力的比较

牙髓封闭剂常规用来封闭牙本质小管,它可以在根充材料和牙本质壁之间创建一层同源界面。然而,生物陶瓷类封闭剂因其生物活性具有更多潜在的特性。经过恰当的牙髓治疗后,骨性愈合极大程度的依赖于成骨细胞的活性和分化。作者推测,EndoSequence BC Sealer (Brasseler, Savannah, GA)和ProRoot ES (Dentsply Tulsa Dental Specialties, Johnson City, TN)与Roth (Roth International, Chicago, IL)和AH Plus (Dentsply DeTrey Gmbh, Konstanz, Germany)相比具有更好的生物相容性和成骨潜力。

研究将1个小鼠成骨前体细胞系(IDG-SW3)暴露于每种不同浓度的封闭剂7天。通过三磷腺苷定量分析(CellTiter-Glo [Promega, Madison, Wisconsin])的冷光检验分析细胞的相对活性。通过对DMP-1表达的荧光显微镜检查、茜素红染色、特异性引物对已知成骨分化标志物(DMP-1、ALP和Phex)的实时反转录聚合酶链式反应检测成骨分化潜力。数据通过2-way analysis of variance或1-way analysis of variance和Bonferroni post hoc检验进行统计学评估。

结果显示,2种生物陶瓷类的封闭剂即使是在高浓度下也具有极优的生物相容性。相反,Roth和AH Plus浓度在低于生物陶瓷组100×时出现了细胞死亡。重要的是,2种生物陶瓷类封闭剂均显著增强成骨分化能力,尽管EndoSequence BC Sealer也出现了较强的反应。2种生物陶瓷类封闭剂组可见DMP-1表达上调,明显升高的成骨分化标志物基因表达和较强的矿化结节形成。而使用Roth或AH Plus时,可见成骨分化和功能显著受到抑制。

结论:EndoSequence BC Sealer和ProRoot ES具有更佳的生物相容性以及促进成骨分化的能力,而AH Plus和Roth的生物活性较差。
