Stroke:脑缺血后进行的miRNAs全基因组揭示再灌注相关的miRNA簇有哪些?

循环中改变的miRNA组与脑缺血/再灌注相关,但与脑出血无关,意味着可以作为急性期再灌注的一个潜在生物学标志物。再灌注诱导的位于X染色体内血清素受体HTR2C内含子中的miR-1264/1298/448集群的病理生理作用需要进一步研究调查。

循环中MicroRNAs(miRNAs),由于其在血液中的高稳定性以及与疾病病理生理相关,正在成为脑卒中的新兴生物学标志物。然而,脑卒中急性期循环中全基因组miRNAs(miRNA组)并没有被全面的进行归纳和总结。近日,卒中领域权威杂志Stroke上发表了一篇研究文章,研究人员针对这一问题进行了研究。

研究人员采用新一代测序方法检测了急性缺血性和出血性脑卒中小鼠模型循坏中的miRNA组。研究人员将脑卒中模型鼠与假手术和正常小鼠相比较来确定循环中表达下降的miRNAs分子,采用定量反转录聚合酶链反应对排名第一的miRNAs进行验证和归纳特征。

与假手术小鼠相比,缺血性脑卒中小鼠3小时后,研究人员发现有24个循环中的miRNAs表达丰度发生了改变,然而在出血性脑卒中小鼠循环中miRNA组并没有改变。在缺血性脑卒中小鼠上调的miRNAs分子中,位于前列的是miR-1264/1298/448集群并且强烈地依赖于不同缺血模型的再灌注情况。通过时程实验表明miR-1264/1298/448集群在循环中再灌注大约3小时后达到高峰,此后逐渐下降。

由此可见,循环中改变的miRNA组与脑缺血/再灌注相关,但与脑出血无关,意味着可以作为急性期再灌注的一个潜在生物学标志物。再灌注诱导的位于X染色体内血清素受体HTR2C内含子中的miR-1264/1298/448集群的病理生理作用需要进一步研究调查。
