SVN:循环内皮微囊泡及其携带的miR-125a-5p可以作为缺血性中风的潜在生物标志物

循环EMVs和EMV-miR-125a-5p与IS的发生、进展、亚型和严重程度密切相关

内皮细胞微泡(EMVs)与内皮细胞(ECs)的状态密切相关。来自我国广东医科的学者早期的研究表明,EMVs可以通过转移其携带的miR-125a-5p在ECs中发挥保护作用。然而,循环的EMV及其携带的miR-125a-5p是否可以作为缺血性中风(IS)的生物标志物仍是未知数。

为此,在本研究中,研究人员招募了72名IS受试者,60名具有高卒中风险的受试者和56名年龄匹配的对照,检测了循环的EMV及其携带的miR-125a-5p(EMV-miR-125a-5p)水平。随后使用微RNA(miR)阵列研究了3名IS患者和3名匹配的健康对照组的血浆EMVs样本中miRs的表达变化。利用短暂性大脑中动脉闭塞(tMCAO)建立IS小鼠模型。

结果显示,IS患者的EMVs水平明显升高,在急性期水平最高,并与颈动脉斑块、颈动脉内膜厚度(IMT)、美国国立卫生研究院卒中量表(NIHSS)和梗死体积呈正相关。相反,观察到EMV-miR-125a-5p水平在IS中明显降低,在急性期水平最低,并与颈动脉斑块、IMT、NIHSS评分、梗塞体积负相关。EMVs和EMV-miR-125a-5p水平与大动脉粥样硬化亚组密切相关。重要的是,EMVs和EMV-miR-125a-5p水平可以作为独立的风险因素,ROC曲线对IS的曲线下面积(AUC)分别达到0.720和0.832,在它们结合后上升到0.881。在IS小鼠模型中,与对照组相比,对照组EMVs或n-EMVs给药可以减少IS小鼠的梗死体积和神经功能缺损评分,同时增加IS小鼠的脑血流量,而IS EMVs或氧和葡萄糖剥夺(OGD)-EMVs给药会加重tMCAO诱发的缺血损伤。此外,我们观察到OGD EMVmiR-125a-5p可以部分改善IS后OGD EMVs引起的脑损伤。

总是,这些发现表明,循环EMVs和EMV-miR-125a-5p与IS的发生、进展、亚型和严重程度密切相关,它们可以作为IS的创新生物标志物和治疗目标,特别是当它们结合在一起时。
