BMC Cancer:膀胱癌免疫微环境相关基因分析,可用于预测患者的预后

浸润性免疫细胞和基质细胞是膀胱癌(BC)微环境的重要组成部分,可显著影响BC的进展和结果。然而,肿瘤浸润免疫细胞的每个亚组的贡献尚不清楚。本研究旨在对肿瘤免疫微环境进行细胞表型和转录分析,分析不同细胞

浸润性免疫细胞和基质细胞是膀胱癌(BC)微环境的重要组成部分,可显著影响BC的进展和结果。然而,肿瘤浸润免疫细胞的每个亚组的贡献尚不清楚。本研究旨在对肿瘤免疫微环境进行细胞表型和转录分析,分析不同细胞亚群和基因与BC预后的关联。

 

从The Cancer Genome Atlas中下载了412例BC患者的临床数据和433个正常和癌症组织的转录文件。采用CIBERSORT算法确定每个样本中22种免疫细胞类型的相对丰度,采用ESTIMATE算法确定BC肿瘤微环境中不同表达的基因,并对其进行功能富集和蛋白-蛋白相互作用(PPI)分析。通过Cox回归分析和Kaplan-Meier方法研究了细胞亚群和不同表达基因与患者生存和临床参数的关联。

 

结果显示,静止的自然杀伤细胞和激活的记忆CD4+和CD8+ T细胞与患者的预后良好相关,而静止的记忆CD4+ T细胞与预后不良相关。差异性表达分析显示,影响免疫和基质细胞评分的基因有1334个;其中97个基因对BC患者的总体生存有预测作用。在PPI网络中前10个有统计学意义的枢纽基因中,CXCL12、FN1、LCK和CXCR4被发现与BC预后相关。

 

综上所述,该研究结果表明,肿瘤浸润免疫细胞和肿瘤微环境相关基因可影响患者的预后,可能是BC患者预后和免疫治疗反应的重要决定因素。
